Bioreducible Zinc (II)-Coordinative Polyethylenimine with Low Molecular Weight for Robust Gene Delivery of Primary and Stem Cells

چکیده
Low molecular weight (LMW) cationic polymers show safety profile and biocompatibility, but are hindered of limited efficacy in gene delivery. This protocol describes a zinc(II) coordinative strategy to transform common LMW highly efficient safe vehicles. exhibit lower compared their high counterparts, largely attributed weaker nucleic acid binding. From this point view, zinc dipicolylamine (Zn-DPA) analogs showing phosphate binding affinity used functionalize obtain higher DNA encapsulation. In addition, for the purpose release, bioreducible disulfide bond is introduced between Zn-DPA analogues, which can be cleaved by abundant glutathione cytoplasm. The Zn coordination dramatically enhances transfection across diverse cell types, including primary stem cells.

Bioreducible polyethylenimine nanoparticles for the efficient delivery of nucleic acids.
Recently, non-viral vectors for nucleic acid delivery have received considerable attention. Among the various non-viral vectors, branched polyethylenimine (bPEI, 25 kDa) has been one of the most widely used carrier systems due to its high transfection efficiency, however, it imparts high cytotoxicity. In this study, we have crosslinked bPEI with a bioreducible linker, 3,3'-dithiodipropionic aci...
متن کاملBiscarbamate cross-linked polyethylenimine derivative with low molecular weight, low cytotoxicity, and high efficiency for gene delivery
Polyethylenimine (PEI), especially PEI 25 kDa, has been widely studied for delivery of nucleic acid drugs both in vitro and in vivo. However, it lacks degradable linkages and is too toxic for therapeutic applications. Hence, low-molecular-weight PEI has been explored as an alternative to PEI 25 kDa. To reduce cytotoxicity and increase transfection efficiency, we designed and synthesized a novel...
متن کاملHigh gene delivery efficiency of alkylated low-molecular-weight polyethylenimine through gemini surfactant-like effect
To our knowledge, the mechanism underlying the high transfection efficiency of alkylated low-molecular-weight polyethylenimine (PEI) is not yet well understood. In this work, we grafted branched PEI (molecular weight of 1,800 Da; bPEI1800) with lauryl chains (C₁₂), and found that bPEI1800-C₁₂ was structurally similar to gemini surfactant and could similarly assemble into micelle-like particles....
متن کاملAmphiphilic graft copolymer based on poly(styrene-co-maleic anhydride) with low molecular weight polyethylenimine for efficient gene delivery
BACKGROUND AND METHODS A new amphiphilic comb-shaped copolymer (SP) was synthesized by conjugating poly(styrene-co-maleic anhydride) with low molecular weight polyethyleneimine for gene delivery. Fourier transform infrared spectrum, (1)H nuclear magnetic resonance, and gel permeation chromatography were used to characterize the graft copolymer. RESULTS The buffering capability of SP was simil...
